Surprised no-one has mentioned Kiefer Sutherland as Jack Bauer in 24... surely one of the most perfect matches of actor and character in our lifetime, you couldn't imagine anyone else in that role.

Another one must be Christopher Reeve (R.I.P.) as Clark Kent/Superman... the fact he still casts a long shadow over that role is testament to just how definitive he was in it.

Someone who was born for a role but never got it is the late Lewis Collins as James Bond, alas...
